True gluten intolerance or Coeliac Disease (CD) affects up to 1% of the EC, US & Australian populations. CD is caused when toxic proteins in cereal gluten damage the absorptive areas of the small intestine. It manifests itself as e.g. intestinal pain, bloating and cramps and in children as a failure to thrive and can only be reversed by strict adherence to a diet low in gluten. Increasingly many people also perceive themselves to be food intolerant and may avoid wheat/gluten as a result.

These phenomena have led to a rise in the availability of premium-priced "gluten free" foods, defined in proposed Codex and EC guidelines as containing less than 100 mg/kg [Parts Per Million; PPM] gluten if cereal derived and less than 20mg/kg gluten for non-cereal derived foods.

Environmental monitoring to detect gluten is increasingly included in gluten control plans within Hazard Analysis Critical Control Point (HACCP) programmes in the food industry. Gluten testing has been brought sharply into focus with the implementation of Directive 2003/89/EC and new Codex Guidelines. Imutest Swabbing/Gluten FlowThrough tests allow swabs to be collected and gluten to be detected in swab samples at very low (sub-ppm) levels quickly and easily and can be used virtually anywhere.

Flocked nylon pile swabs, specifically designed to pick up and quickly shed proteins, are immersed in a specially formulated swabbing solution then used to swab a representative area of surface, hard-to-reach location (e.g. within complex equipment), air sampling device etc. When the swab is replaced in the tube it can be snapped off and securely capped.

The solution in the tube (approx. 2mls) helps dissolve and stabilise any gluten present and is used to carry out a Gluten FlowThrough test immediately to check for the presence of gluten in the swabbing solution.

The swabbing solution is added directly to the GFT test card and allowed to percolate into the card (2-3 minutes); when it has drained through, the test area is wiped and the visualisation reagent added (colour develops in 2-3 minutes as the reagent passes through).

As the visualisation reagent drains into the test card, a pattern of spots appears:

  • One spot on the right hand side:       NEGATIVE for gluten (less than 0.5ppm)
  • Two spots in the test area:                POSITIVE for gluten

 Each self-contained kit comes ready to use and comprises:

  • 5, 10 or 20 individually pouched, sterile flocked nylon pile swabs
  • 5, 10 or 20 pre-filled tubes of stabilising & preservative Swabbing Solution
  • 5, 10 or 20 Sets of GFT Test Cards, Visualisation Reagents and other required parts
  • Instructions For Use

The swabbing solution is a buffered saline solution with food grade surfactant, a vivid blue food dye to help visualise the swabbed area and facilitate cleaning afterwards and a non-toxic preservative. The kits are therefore completely safe to handle and use for food applications and virtually everything you need is in the box.

How to take a swab sample:

  1. Ensure that hands & clothing are clean and will not contaminate swabbed samples; it is advisable to wear gloves and wash them between swabs.
  2. Using an indelible marker, label a Swabbing Solution tube with an identifying code.
  3. Remove a swab from its pouch by carefully pushing the non-flocked end of the shaft through the film and withdrawing the swab through the hole created.
  4. Remove the cap from the labelled swabbing tube and dip the swab in the liquid.
  5. Take out the swab by sliding the tip up the inside of the tube to remove excess solution.
  6. Use the wetted tip to swab thoroughly an area of e.g. 5x5cm or 10x10cm in an orderly pattern; this is often done by criss-crossing the area from one side to the other while rotating the swab shaft, then repeating at 90° to the first strokes.
  7. Place the swab back in the tube, rotate the shaft with the tip against the side of the tube to mix, then withdraw and repeat the swabbing process described above.
  8. Place the swab back in the tube and carefully break off the thinner part of the shaft nearer the flocked tip. This is most easily done by replacing the swab in the tube, partially replacing the cap, trapping the shaft between the cap and the top rim of the tube, then carefully bending the shaft to a right angle against the rim of the tube.
  9. Re-cap the tube and mix well to disperse the swabbed material in the solution.

To test for gluten using the GFT test components:

  • Fill the GFT test card well with swabbing solution using the pipetteprovided and allow it to percolate into the card; when it has drained through, wipe the test area using the cotton wipe provided.
  • Add the visualisation reagent added to the test well and allow to drain into the card.

As this reagent drains into the test card, a pattern of spots appears:

  • One spot on the right hand side:       NEGATIVE for gluten (less than 0.5ppm)
  • Two spots in the test area:               POSITIVE for gluten

The swabbing and testing process can be completed in less than 15 minutes and can be done almost anywhere.
