Sesame-Check kits quantitatively detect sesame seeds in fresh & processed food products and surface swabs. The kits use a simple 3-step, 60 minute sandwich assay format and contain all the reagents required for the extraction and detection of sesame between 2 & 20 mg/kg (PPM); the LOD is 0.2 PPM sesame. All allergen-specific reagents are ready to use and the kits are easily automated.

The assay utilises polyclonal antibodies to detect sesame proteins present in extracts of food-related samples containing sesame.
The assay is a direct “sandwich” ELISA, designed to detect sesame proteins at very low levels in fresh, processed and heated food products as well as in environmental swabs.
The assay range is nominally between 2.0-20 ppm (mg/kg) sesame seeds if the stated extraction ratio is used and the sample extract is not diluted. The range can be extended upwards by amending the extraction ratio, and/or diluting the sample extract, if required. This range corresponds to 0.4-4ppm sesame protein.
The Limit Of Detection (LOD) of the assay is ~0.2 ppm (mg/kg) sesame seeds, which corresponds to 0.04ppm sesame protein.

Laboratory Sample Preparation
- Prepare Sample by grinding/chopping/blending until homogeneous.
- Add 1 part Test Portion to 20 parts Extraction Buffer 1X.
- Extract Incubate for 15 minutes @ 60oC Shaking every two minutes.
- Separate Centrifuge for 10 minutes.
- The sample extract can be used in several other "Allergen-Check" ELISAs.
ELISA Protocol
- Pipette 100µL Standards and Sample Extracts into wells.
- Mix. Incubate at room temperature for 20 minutes.
- Wash wells three times with Wash Solution.
- Pipette 100µL HRP-Conjugate into wells.
- Mix. Incubate at room temperature for 20 minutes.
- Wash wells three times with Wash Solution.
- Pipette 100µL TMB Substrate reagent into wells.
- Mix. Incubate at room temperature in the dark for 20 minutes.
- Pipette 100µL Acid Stop Solution into wells.
- Mix. Read wells at 450nm wavelength within 15 minutes.
- Calculate PPM Sesame results for all Samples.
