Hazelnut-Check kits quantitatively detect hazelnuts in fresh & processed food products and surface swabs. The kits use a simple 3-step, 60 minute sandwich assay format and contain all the reagents required for the extraction and detection of hazelnut between 1 & 40 mg/kg (PPM); the LOD is <0.5 PPM of hazelnut. All allergen-specific reagents are ready to use and the kits are easily automated.

The assay utilises polyclonal antibodies to detect the mixture of proteins present in soluble extracts of hazelnut using a direct “sandwich” ELISA technique.

It is designed to detect hazelnut at very low levels in fresh, processed and heated food products as well as in environmental swabs.

The assay range is nominally between 1-40 mg/kg (PPM) hazelnut, which corresponds to ~0.15-6.0ppm hazelnut protein if the stated extraction ratio is used and samples are not diluted. The range can be extended upwards by amending these ratios if required.

The Limit Of Detection (LOD) of the assay is <0.5PPM hazelnut, which corresponds to <0.1 PPM hazelnut protein.

Laboratory Sample Preparation

  • Prepare Sample by grinding/chopping/blending until homogeneous.
  • Add 1 part Test Portion to 20 parts Extraction Buffer 1X.
  • Extract Incubate for 15 minutes @ 60°C Shaking every 2 minutes.
  • Separate Centrifuge for 10 minutes.
  • The sample extract can be used in several other "Allergen-Check" ELISAs.

ELISA Protocol

  • Pipette 100µL Standards and Sample Extracts into wells.
  • Mix. Incubate at room temperature for 20 minutes.
  • Wash wells three times with Wash Solution.
  • Pipette 100µL HRP-Conjugate into wells.
  • Mix. Incubate at room temperature for 20 minutes.
  • Wash wells three times with Wash Solution.
  • Pipette 100µL TMB Substrate reagent into wells.
  • Mix. Incubate at room temperature in the dark for 20 minutes.
  • Pipette 100µL Acid Stop Solution into wells.
  • Mix. Read wells at 450nm wavelength within 15 minutes.
  • Calculate PPM Hazelnut protein results for all Samples.
