True gluten intolerance or Coeliac Disease (CD) affects 1% or more of the EC & US populations; it is caused when toxic proteins in cereal gluten damage the absorptive areas of the small intestine. CD manifests itself as e.g. intestinal pain, bloating and cramps, and in children as a failure to thrive. CD can only be reversed by strict adherence to a diet low in gluten.
Increasingly many people also perceive themselves to be food intolerant and may avoid wheat/gluten as a result. These phenomena have led to a rise in the availability of “gluten-free” food products and the consequent requirement to monitor gluten levels in such products more closely.
Codex Standard 118 and recent EC Regulations define “gluten free” foods for PARticular NUTritional use (PARNUTS) as containing less than 20 Parts Per Million (PPM or mg/kg). PARNUTS foods above 20 PPM but below 100 PPM must be labelled “very low gluten”.
D.I.L.’s Imutest GLUTEN FlowThrough (GFT) kits are fast and sensitive tests for the presence of cereal gluten in food raw materials, rinse waters, finished products etc. The test is very simple to use and requires no additional equipment; it is designed for use in food factories with limited testing facilities, small laboratories with lower testing volumes and can even be used at home. Several food samples can be tested within 20 minutes and gluten levels of 10-20 parts per million (ppm) detected.
GFT tests are ideal for powders/flours, fine crumbs, liquids/sauces etc, which need no further preparation as they should be homogeneous and e.g. biscuits that are usually easily crushed.
If necessary though, food samples need to be prepared e.g. by grinding/chopping etc to make them homogeneous and ready for testing.
Any gluten in the sample is then dissolved using a highly effective extraction solution that maximises gluten recovery.
After the extraction mixture has settled a few drops are added to a Diluent Solution and mixed.
This solution is added to the GFT test card and allowed to percolate into the card; when it has drained through, the test area is wiped and the visualisation reagent added.
As this reagent drains into the test card, a pattern of spots appears:
- One spot on the right hand side: NEGATIVE for gluten (less than 0.5ppm)
- Two spots in the test area: POSITIVE for gluten
- If necessary, prepare your food sample so that it is homogeneous and ready for extraction and testing
- Weigh out, or estimate using the spoons provided, the amount of food required (approx. 0.5G)
- Add the food sample to a pre-filled tube of Extraction Solution; mix well; allow to stand; mix again. (The application of heat e.g. by dropping the tube into hot tap water improves extraction efficiency)
- Allow this mixture to settle out until a relatively clear layer appears between any froth at the top and settled food debris below.
- Using the pipette provided, add a few drops of this settled extract to a pre-filled tube of Diluent Solution; mix well.
- Add the dliuted food sample extract to the GFT test card well and allow it to percolate into the card; when it has drained through, wipe the test area using the cotton wipe provided.
- Add the visualisation reagent added to the test well and allow to drain into the card.
As this reagent drains into the test card, a pattern of spots appears:
- One spot on the right hand side: NEGATIVE for gluten (less than about 10-20ppm)
- Two spots in the test area: POSITIVE for gluten
The extraction and testing process can be completed in about 15-20 minutes and can be done almost anywhere.
GFT 50 test sets contain 50 sets of all GFT test components, bulk packed for larger users.
GFT test sets (No Plastics) contain 20 bulk packed tubes of food sample extraction solution and diluent liquid, test cards and visualisation reagents but with no additional plastic consumables.
An ideal version for laboratory use, as tests can be completed in 10-20 minutes but do require some consumables, which are normally available in most small laboratories.
