β-LG-Check kits quantitatively detect β-Lactoglobulin in fresh and lightly-processed food products and surface swabs. They use a simple, 3-step, 60 minute sandwich assay format and contain all the reagents required for the extraction and detection of skim milk powder (SMP) between 0.2 & 7.0 mg/kg (PPM); the LOD is <0.05 PPM of SMP. All allergen-specific reagents are ready to use and the kits are easily automated.

Skim Milk Powder (SMP) contains approximately 4.5% β-Lactoglobulin (β-LG), which has been identified as a significant allergen.

This assay utilises polyclonal antibodies to detect β-LG, as a marker for the presence of milk and/or whey proteins, using a direct “sandwich” ELISA technique.

It is designed to detect β-LG at low levels in food raw materials, fresh and lightly processed food products (β-LG is relatively unstable to heat above ~60°C) as well as in environmental swabs.

The assay range is nominally between ~10-300 mg/kg (PPB) β-LG, which corresponds to ~0.2-7.0 PPM SMP content if the stated extraction ratio is used and the sample is not diluted. The range can be extended upwards by amending the extraction ratio and/or diluting the sample extract if required.

The Limit Of Detection (LOD) of the assay is ~1.5 mg/kg (PPB) β-LG, which corresponds to <0.05PPM SMP content.

Laboratory Sample Preparation

  • Prepare Sample by grinding/chopping/blending until homogeneous.
  • Add 1 part Test Portion to 20 parts Extraction Buffer 1X.
  • Extract Incubate for 15 minutes @ 60oC Shaking every two minutes.
  • Separate Centrifuge for 10 minutes.
  • The sample extract can be used in several other "Allergen-Check" ELISAs.

ELISA Protocol

  • Pipette 100µL Standards and Sample Extracts into wells.
  • Mix. Incubate at room temperature for 20 minutes.
  • Wash wells three times with Wash Solution.
  • Pipette 100µL working HRP-Conjugate into wells.
  • Mix. Incubate at room temperature for 20 minutes.
  • Wash wells three times with Wash Solution.
  • Pipette 100µL TMB Substrate reagent into wells.
  • Mix. Incubate at room temperature in the dark for 20 minutes.
  • Pipette 100µL Acid Stop Solution 1 into wells.
  • Mix. Read wells at 450nm wavelength within 15 minutes.
  • Calculate PPM SMP results for all Samples.
